Abstract:

To provide a method for performing real-time polymerase chain reaction capable of accurately determining the amplified nucleic acid quantity of a target nucleic acid fragment formed by PCR, a real-time polymerase chain reaction apparatus for the method for performing real-time polymerase chain reaction, and a pyrophosphate sensor electrode.

The method for performing real-time polymerase chain reaction is characterized by a polymerase chain reaction by treating a target nucleic acid fragment with a polymerase in the presence of a primer pair while electrochemically measuring the concentration of pyrophosphoric acid by-produced in the amplification step of the target nucleic acid fragment. The concentration of pyrophosphoric acid is determined by a pyrophosphate sensor electrode to detect phosphate anion (PO43-) by the by-produced pyrophosphoric acid in the real-time polymerase chain reaction.
