Abstract:

PURPOSE: To produce by a specific method a novel saccharide oligopeptide derivative capable of atereo selectively and effectively giving a saccharide oligopeptide useful for the clarification of the receptor structure of lectin for recognizing sialosyl Tn epitope.
CONSTITUTION: A compound of formula I (R is an OH-protecting group; X is halogen) is condensed with a compound of formula II (R1 is H, methyl; R2 is phenacyl, allyl) preferably in methylene chloride at -15 to 5°C for 3-5hr. The condensation product is usually subjected to a benzylidene group-removing reaction and subsequently reacted with a compound of formula III (R3 is an OHprotecting group; R4 is a COOH-protecting group; Y is halogen) preferably in a solvent such as carbon tetrachloride at -5 to 25°C for 10-16hr. The condensation product is successively subjected to an acetoamide-forming reaction, an allyl group-removing reaction and a thiophenyl group-removing reaction to produce the objective saccharide oligopeptide derivative of formula IV (Z is H, thiophenyl).
