Abstract:
PURPOSE: To provide an enzyme donor polypeptide and an enzyme acceptor polypeptide used for an enzyme complementarity assay for a quantitative analysis of specimens of both high molecular weight and low molecular weight at a high sensitivity.
CONSTITUTION: The existence of dubious specimens in a sample is detected by a method comprising the following steps: (a) a reaction mixture is formed by combining a medium a specimen, an enzyme donor polypeptide bonded body, a specimen honding protein, an enzyme acceptor polypeptide and a substrate; (b) the invert ratio of the substrate in the reaction mixture is measured; (c) the amount of the specimens in the sample is measured by comparing the invert ratio of the substrate with an invert ratio of the substrate obtained by using a specimen of known concentration. Furthermore, an improved method for an enzyme complementarity assay for measuring the amount of specimens comprises a step (d) in which at least one enzyme donor or enzyme acceptor polypeptide prepeared by changing the amino acid sequence in order to increase the association constant of the enzyme donor bonded body and the enzyme acceptor is used.
