To provide a simple method for separating/recovering a microorganism existing in activated sludge used for treating metal-containing wastewater, a method for detecting the microorganism with which a DNA sequence can be read without inhibiting an enzymic reaction such as a polymerase chain reaction (PCR) and a monitoring technique for the microorganism in the activated sludge used for treating the metal-containing wastewater.
The method for recovering the microorganism from the activated sludge used for treating the metal-containing wastewater is characterized as follows. The activated sludge used for treating the metal-containing wastewater is regulated to pH≥7 and ≤10 and metal ions in the activated sludge are converted into a metal hydroxide. The activated sludge is then allowed to stand for a prescribed time to precipitate the metal hydroxide. The resultant supernatant is further filtered through a membrane having ≤0.5 μm pore diameter to separate the microorganism in the activated sludge on the filter membrane. The method for identifying the microorganism in the activated sludge used for treating the metal-containing wastewater is characterized as follows. The DNA of the recovered microorganism is extracted and a specific region of the DNA is then amplified to identify the microorganism based on the DNA sequence of the resultant DNA product.
