Abstract:

PURPOSE: To provide the method for removing the cellular wall polysaccharide of Porphyridium curentum, capable of rapidly and readily convert the Porphyridium curentum into its protoplast.

CONSTITUTION: This method for removing the cellular wall polysaccharide of the Porphyridium curentum to convert the Porphyridium curentum into its protoplast or spheroplast is characterized by employing the enzyme mixture of pectinase and amylase or the enzyme mixture of the pectinase and cellulase. The cellular wall polysaccharide of the Porphyridium curentum can efficiently and rapidly removed to provide its protoplast or spheroplast. Thereby, the method permits to isolate the bred cells high in polysaccharide productivity, or to convert the cells into the protoplasts and subsequently fuse the protoplasts into a new cell further higher in the polysaccharide productivity. Essential genes can be supplied by the breeding of the cells.
