PURPOSE: To make it possible to isolate individual protoplasts under a microscope using a fluorescent dyestuff as an index, by dyeing the protoplasts of basidiomycetes with the fluorescent dyestuff, and fusing the protoplasts.
CONSTITUTION: Cell walls of two species of basidiomycetes of different species, genuses, families or orders or two strains of the same species are dissolved with a cell wall lytic enzyme to form protoplasts, which are then suspended in a solution of a fluorescent dyestuff, e.g. 4,6-diamidio-2-phenylindole, to dye either one or both colors of the protoplasts to be fused. The unreacted fluorescent dyestuff is then completely removed, and fusion treatment is carried out using a fusion solution containing polyethylene glycol, etc. to make it possible to isolate the individual fused protoplasts under a microscope using the fluorescent dyestuff as an index.
