To provide a method for producing a polypeptide mixture labeled with a stable isotope element utilizable in an improved method for NMR measurement of a protein.
The method for producing the polypeptide mixture labeled with the stable isotope element comprises the following steps. (1) a step of determining an amino acid sequence X1-...-Xn of a peptide composed of n amino acid residues, (2) a step of preparing amino acid cocktails for synthesis x1, x2, ... and xn corresponding to each of the amino acids X1, X2, ... and Xn, respectively and (3) a step of synthesizing a peptide composed of the amino acid sequence determined in (1) using the amino acid cocktails for synthesis prepared in (2). Thereby, the polypeptide remarkably simplifying measuring operation of the protein by NMR and providing the new method for NMR measurement of the protein can be produced without the need of preparing the polypeptide for the number of the amino acid residues while changing the amino acid residues to be labeled as opposed to the conventional method.
