Abstract:
To provide a use of a composition for treatment of a hereditary disease.SOLUTION: Provided is a use of a composition that includes a target construct including a target sequence with an inverse orientation to an exogenous DNA sequence, a complemental strand oligonucleotide homologous to the target sequence, and CRISPR nuclease, where the exogenous DNA sequence includes a difference from at least one nucleotide comparing to a genome, a target sequence and a target nucleic acid sequence in the genome are recognized by CRISPR nuclease, the target nucleic acid sequence in the genome, when the exogenous DNA sequence is once incorporated into the genome of a non-dividing cell in an accurate orientation, is not present any more in the genome.SELECTED DRAWING: Figure 1F
