Abstract:
PROBLEM TO BE SOLVED: To provide, in part, endpoint PCR assays for the detection and high throughput zygosity analysis of the fad-2 gene in canola.SOLUTION: Disclosed herein is a method for determining the zygosity of a canola plant having fad-2 gene, comprising a step in which a genomic DNA sample is taken from a canola plant; a step in which an amplicon is formed; a step in which a fluorescent probe preferentially forms a hybrid with the amplicon; and a step of showing that the sample contains a first allele with a single nucleotide polymorphism of interest and a second allele without the single nucleotide polymorphism of interest. Further disclosed is a related kit for determining zygosity from a sample of a canola plant or seed which can be used to uniquely identify the zygosity and variety of canola lines comprising the subject gene.SELECTED DRAWING: None
