Abstract:
PROBLEM TO BE SOLVED: To provide a method for remodeling and labeling proteins and antibodies by oligosaccharide conjugation, and to provide antibodies or proteins that bind a modified oligosaccharide.SOLUTION: The method of the invention comprises the steps of: conjugating a chemical handle comprising a modified sugar to a GlcNAc residue on a first protein; and mixing the first protein with a reporter molecule, carrier molecule or solid support that is reactive with the chemical handle. The reporter molecule, carrier molecule or solid support binds to the protein at the position of the chemical handle to form a modified glycoprotein. In certain embodiments, the first protein is an antibody.
