Abstract:
To provide a modified cDNA that expresses the factor VIII or derivative thereof at a high level.
The cDNA is obtained by introducing the nucleotide sequence that can be spliced one or more times or a nucleotide sequence that is spliced while the pre-mRNA is transferred from the cell nucleus in the intron 1 and/or the intron 13 of the cDNA of the wind type factor VIII. The animal cells including the recombinant expression vector including the modified factor VIII cDNA are cultured to achieve the object of the invention. The transfer vector to be used for human gene therapy including the modified factor VIII cDNA is disclosed.
