To provide a novel modified nucleotide which contains a nucleotide prepared by introducing a detectable signaling zone into a naturally occurring nucleotide and enables stable, economical, prompt and reproducible detection of a nucleic acid as a replacement for a radioactive probe.
This modified nucleotide is an oligo- or polynucleotide containing a modified nucleotide represented by formula I (B is purine covalently bonding to the C1'-position of a saccharide moiety, 7-deazapurine, pyrimidine, and when B is purine or 7-deazapurine, the saccharide moiety bonds to the N9-positions; when B is pyrimidine, the saccharide moiety bonds to the N1-position of the pyrimidine; A is at least one component which has 3 or more carbon atoms and is a signaling zone that can form a detectable signal; B and A are directly linked through a covalent bond or through a linking group; one of (x) and (y) represents a group of formula II or formula III and the other is nothing or OH, H, in the case of a terminal nucleotide) and is useful as a non- radioactive probe.
