NEW MATERIAL:A modified oligodeoxynucleotide containing deoxy-7,8- dihydroxyadenosin-8-one.
USE: An inhibitor of nuclease, ligand in purifying nuclease with an affinity chromatography, etc. The above-mentioned new material has high resistance to the nuclease.
PREPARATION: A deoxy-N-acetyl-7,8-dihydroadenosin-8-one expressed by formula I is reacted with dimethoxytrityl chloride to protect 5 ' position of the compound expressed by formula I and the protected compound expressed by formula I is hydrolyzed in the presence of a catalytic amount of pyridine in a solvent such as dichloromethane by [(F3C)2CHO]3P. Then the resultant compound expressed by formula II (DMTr is dimethoxytrityl) is polymerized with other nucleotide according to an ordinary method and then the product is successively treated with dichloroacetic acid and ammonia water to deprotect the product.
