PURPOSE: To obtain a new antibody specifically reacting with a ribonucleotide reductase(RNR) R2 subunit, having a RNR activity-inhibiting action, and capable of detecting the RNR for cell cycle markers on the diagnosis of tissues, etc.
CONSTITUTION: The C-terminal peptide of a human ribonucleotide reductase(RNR) R2 subunit having an amino acid sequence of the formula is synthesized, and bound to keyhole limpet hemocyanin(KLH). The produced conjugate is administered as an immunogen into a rat to immunize the rat. After the immunization is finished, splenic cells are collected from the immunized rat, fused with myeloma cells, and subsequently selectively cultured in a HAT culture medium. The obtained hybridoma cells are screened to select an antibody- producing strain. The stain is cloned and subsequently cultured to obtain the subject new antibody specifically reacting with the human RNR R2 subunit, having an action for inhibiting the activity of the RNR and useful as a cell cycle marker, etc., on the diagnosis of tissues, the diagnosis of cells, etc.
