PURPOSE: To obtain a new monoclonal antibody having strong reactivity to a main adventitial protein and basic small body of Chlamydia pneumoniae and useful as a diagnostic for detecting Chlamydia pneumoniae, a reagent, a medicine, etc.
CONSTITUTION: Human lung-derived cell is infected with Chlamydia pneumoniae YK-41 strain and the strain is cultured and the lung-derived cell is separated and crushed and centrifuged and the supernatant is collected to provide a basic small body solution of Chlamydia pneumoniae. Then, the solution is separated and purified by disc preparative electrophoresis to prepare main adventitial proteins and the proteins are mixed with complete Freund's adjuvant and the mixture is fused with a mouse myeloma cell and the fused cell is selectively collected in HAT culture medium to provide a hybridoma, which is screened and subjected to cloning by a limiting dilution analysis and cultured to provide the objective new monoclonal antibody capable of reacting a main adventitial protein and the main small body of Chlamydia pneumoniae.
