Abstract:
PURPOSE: To obtain a new compound usuful as a drug effective as hypolipidemic medicine with a small amount which suppress the bile acid absorption concentration-dependently and antagonistically, and has high affinity for the peculiar bile acid transportation system of small intestine.
CONSTITUTION: The monomeric bile acid derivatives of formula Z-X-GS [GS is a bile acid group which has acid functional receptor on a side chain or its salt; X is a covalent bond, crosslinking group of formula (CH2)n, (n=1-10), alkylen chain is 1 to 3 oxygen which may have NH or NHC(=O); Z is HO-, CH3-O-, HO-CH2-CH=CH-CH2- or the like], for example, the compound of formula I (n=5), is obtained by to dissolving a methylesther of formula II into tetrahydrofuran or 1,4-dioxane, stirring hard with 2N-NaOH at room temperature. It is preferable that Gs is linked to X at third position and the linkage is taken place at α- or β position.
