Abstract:

NEW MATERIAL:The mutant human prourokinase (human PUK) modified in such a manner as to include an amino acid sequence of formula Asn-X-Y (X is amino acid residue; Y is Ser or Thr) in an epidermal growth factor(EGF) domain of human PUK.

USE: A fibrinolytic enzyme having thrombolytic activity. It has long half-life in blood similar to that of a mutant human PUK free from EGF domain. It has high affinity to fibrin and extremely low bleeding tendency.

PREPARATION: A DNA sequence coding a mutant human PUK modified to introduce an amino acid residue of formula Asn-X-Y into the EGF domain of human PUK is integrated in a plasmid and a host transformed with the plasmid is subjected to manifestation.
