Abstract:

PURPOSE: To separate nucleotide useful as a raw material for drugs, etc. from nucleoside, by bringing a solution containing nucleoside and nucleotide at specific pH into contact with active carbon so that both are adsorbed on it, followed by eluting nucleotide with an aqueous solution of an alkali metal hydroxide.

CONSTITUTION: A solution containing nucleoside (e.g., inosine, etc.) and nucleotide(e.g., inosine-5-phosphate, etc.) adjusted to about ≤7.0 pH is passed over a column packed with active carbon, nucleoside and nucleotide are adsorbed on the active carbon, and nucleotide is eluted with an aqueous solution of an alkali metal hydroxide (e.g., sodium hydroxide, etc.). An aqueous solution containing the alkali metal hydroxide and a lower alcohol is passed over the active carbon column from which nucleotide is eluted, and nucleoside is eluted, so that nucleotide and nucleotide and nucleoside are separated mutually.
