PURPOSE: To obtain the subject new compound which is an N-substituted carbamide derivative of 3-carboxy-androst-3,5-diene and useful for treatment, etc., of acne, seborrhoica, hirsutism, alopecia, etc., as a testosterone 5α-reductase inhibitor.
CONSTITUTION: This new N-substituted carbamide derivative of 3-carboxy- androsto-3,5-diene is represented by formula I [R is a hydrocarbon group such as a (substituted) 1- or 2-adamantyl or 1, 2- or 7-norbornanyl 17β]. The compound has testosterone 5α-reductase inhibiting activity and topically useful in treatment of acne, seborrhea, female hirsutism, male alopecia, etc., and systemically in treatment of benign prostatic hypertrophy. The compound is obtained by reacting a steroid acid of formula II with trifluoromethylsulfonyl anhydride, activating carboxylic group of the resultant material and reacting carboxylic group of the material with admantylamine and reacting the reaction product with carbon monoxide in the presence of Pd catalyst to carry out carbonylation.
