Abstract:

PROBLEM TO BE SOLVED: To obtain the subject compound without requiring separation of recemic compounds, by asymmetrically hydrogenating a specific compound in the presence of a complex composed of an optically active diphosphine ligand and indium.
SOLUTION: (A) A compound of formula I (HX is a mineral acid selected from among HBF4, H2, SO4, HPF6, HBr, HI, HCl HSbF6 and HClO4 or a strong organic acid selected from among a 1-8C alkyl SO3H, picric acid, formic acid, a lower alkylcarboxylic acid, an arylcarboxylic acid and a dicarboxylic acid) is asymmetrically hydrogenated in the presence of (B) a complex composed of an optically active disphosphine ligand and indium and optionally (C) a base at 10-200°C under 1-250 bar to give an (R)- or (S)-1-(4- methoxybenzyl)-1,2,3,4,5,6,7,8-octahydroisoquinoline adduct of formula II. The molar ratio of the component A is the component B is preferably 20-80,000.
