Abstract:
PURPOSE: To obtain the subject delivery controlling composition capable of controlling the delivery of an effecting medicine, useful as a medicine for a rectal administration, and consisting of a delivery controlling matrix containing sodium alginate, a calcium salt, the effecting medicine and an excipient dissolvable in a liquid in the rectum.
CONSTITUTION: This medicinal delivery controlling composition is obtained by preparing a delivery controlling matrix by mixing sodium alginate, a calcium salt capable of forming crosslinks with the sodium alginate on exposing the matrix to an aqueous solution and a higher aliphatic alcohol, adding an effective medicine (e.g. amitriptyline) to the matrix in an effective amount capable of suspending or distributing in the matrix, and further adding a sufficient amount of the calcium salt for controlling the delivery of the effective medicine from the matrix on exposing the matrix to the aqueous solution and an excipient melting or dissolving in the liquid of the rectum in the case of a rectal administration, or a higher aliphatic alcohol in the case of an oral administration.
