Abstract:

PURPOSE: To provide a method for easily and accurately determining the quantity of endotoxin in a liposome dispersion.

CONSTITUTION: The quantity of endotoxin in a liposome dispersion is determined by a synthetic substrate method by adjusting the lipid concentration of the liposome dispersion to ≤4.0g/dl, measuring the absorbances Abs (treated) of the prepared specimen treated with a reaction reagent, Abs (untreated) of the specimen treated with water, Abs (blank) of the blank specimen and Abs(standard) of the standard liquid and calculating the concentration (C) of endotoxin in the liposome dispersion from the measured values by the formula, C= Et×{[Abs (treated)-Abs (untreated)-Abs (blank)]/[Abs (standard)Abs (blank)]}.
