Abstract:
PURPOSE: To obtain the subject compound useful for treating ovarian, mammary, pulmonary cancers, etc., with good productivity by culturing a tissue, a cell, etc., of a plant capable of producing a taxane type diterpene using a gas containing oxygen and further gaseous carbon dioxide at a specific concentration.
CONSTITUTION: A tissue (e.g. a stem) of a plant (e.g. Taxus breviflolia) capable of producing a taxane type diterpene (e.g. taxol) is bedded on a solid culture medium containing gellan gum, etc., to carry out the stationary culture in the dark at 25°C. A callus is produced and the resultant cultured cell is transferred to a liquid culture medium and cultured at 25°C in the dark for 21 days under stirring while aerating a gas containing oxygen and further gaseous carbon dioxide at ≥0.04% concentration. The cell is then recovered by filtration and the recovered cell is freeze-dried to carry out the extraction with methanol, etc. The solvent is distilled away from the extract solution and the purification is performed according to the high-performance liquid chromatography. Thereby, the objective taxane type diterpene useful as a therapeutic agent for ovarian, mammary, pulmonary cancers, etc., is obtained with improved productivity.
