Abstract:
PURPOSE: To obtain a novel compound good in crystallinity, useful as a starting material for producing strong active gestagen used for contraceptive regulators and capable of producing gestoden by one shot method in good yield.
CONSTITUTION: This 3-alkoxy-18-methyl-3,5-eatradine-17-one is a compound expressed by formula I [R is a 1-3C alkyl; A-B is HC16=C15H or formula II (R' is a ≤10C alkyl)], e.g. 3-methoxy-18-methyl-3,5,15-estratrien-17-one. The compound is obtained, e.g. by reacting 15α-hydroxyl-18-methyl-4-estren-3,17-dione with methanesulfonyl chloride in pyridine to obtain its 15α-mesylate, reacting the 15α-mesylate with anhydrous sodium acetate in DMF to dehydrate from 15, 16 sites of the 15α-hydroxy-18-methyl-4-estren-3,17-dione and reacting the produced 18-methyl-4,15-estradien-3,17-diene with p-toluenesulfonic acid.
