Abstract:

PURPOSE: To efficiently form mycorrhizal fungi such as Trichola matsutake Sing. or Lyophyllum shimeji Hongo by inoculating cultured mycelia of the mycorrhizal fungi into a raw root of a parasitic plant, then embedding the inoculated part in a sterilized gel, artificially rooting the mycorrhizal fungi and preventing various germs from propagating.
CONSTITUTION: A raw root of a parasitic plant such as Japanese red pine about 25 years old in which fine roots were developed by a method for excavating and digging around the roots for transplanting the year before last is dug out and soil is washed away with river water. The root is then sterilized with a 70% alcohol and a 5% solution of sodium hypochlorite and sufficiently washed with sterile water. The resultant washed root is placed in a sterilized container made of polypropylene and a mixture solution of mycelia prepared by mixing cultured mycelia of mycorrhizal fungi such as Trichola matsutake Sing. or Lyophyllum shimeji Hongo with a 2% solution of sodium alginate is further placed. A sterilized 50mM solution of calcium chloride is added to gelatinize the mixture solution. The root embedded in the sterilized gel is then taken out of the container, covered with MASA soil thereon and cultured to efficiently form the mycorrhiza of the mycorrhizal fungi.
