Abstract:

PROBLEM TO BE SOLVED: To obtain the subject new enzyme for e.g. cleaning composition, consisting of alkaline pectic lyase which scissors a α-1, 4 bond of pectic acid by β-elimination reaction and produces e.g. digalacturonide having a double bond at its unreduced terminal.
SOLUTION: This new alkaline pectic acid lyase is to has such a function as to endo-fashion scissor α-1, 4 bond of pectic acid (polygalacturonic acid) by β-elimination reaction and provide an unreduced C4-C5 site with a double bond to form unsaturated digalacturonide or unsaturated oligogalacturonide, and has a molecular weight of about 20 to 21 kDa (ultracentrifugal equilibrium sedimentation method) in a pH of 5 to 11.5 (for 60 minutes in a 40 mM Britton- Robinson universal buffer solution at 30°C), wherein the optimum pH of reaction is 10.3 to 10.7 (in a 50 mM glycine-sodium hydroxide buffer solution, using pectin as a substrate). This enzyme is useful for a cleaning composition for clothes and dishes, and can be obtained from Bacillus sp. KSM-P15 (FERM P-15987).
