Abstract:
PURPOSE: To control nematodes by bringing a toxin produced by a Bacillus thuringiensis gene product clone pMVC-383 into contact with the nematodes. CONSTITUTION: An ECoRI-HindIII fragment of about 3.4kb containing a code region of a Bacillus thuringiensis variety israelensis (B. t. i.) toxic gene of aboout 130kDa from a B. t. i. strain HD567 is cloned in an ECoRI HindIII position of an expression vector pKK223.3 containing a tac promoter. A 32-mer synthetic oligonucleotide (A) is inserted into the EcoRI position in order to restore an ATG initiation codon and introduce a Shine-Dalgarno(SD) region thereinto. Thereby, a plasmid pMYC.383 is obtained and then transferred into Escherichia coli JM101 to produce a toxin. The toxic gene is genetically integrated into a plant or another microorganism capable of surviving and proliferating in the plant to control soil nematodes and plant parasites. Otherwise, the toxin is administered to an animal host to control the nematodes.
