Abstract:
PURPOSE: To obtain the subject enzyme preparation capable of efficiently extracting astaxanthin in the cells of a yeast, Phaffia rhodozyma, by culturing a new Streptomyces stain producing an enzyme capable of lysing the cell wall of the yeast in a medium containing an inducing substrate.
CONSTITUTION: The new microorganism, Streptomyces rochei PHA-34 strain (FERM P-14284), producing an enzyme capable of lysing the cell wall of an enzyme, Phaffia rhodozyma, is obtained by screening Phaffia rhodozyma cell wall-lysing enzyme-producing fungi from a soil or a leaf soil, and subsequently selecting one stain of Streptomyces rochei highest in the activity and having a wide lysis spectrum capable of lysing also the yeasts of Basidiomycetes and the yeasts of imperfect fungi. The new strain is subsequently aerobically cultured in a liquid medium containing an inducing substrate and used for the production of the enzyme and subsequently recovering the cell wall-lysing enzyme from the culture solution to obtain the enzyme preparation containing the cell wall- lysing enzyme capable of efficiently extracting the astaxanthin in the cells of the yeast.
