Abstract:

PROBLEM TO BE SOLVED: To obtain a DNA useful for preparing a trasnformant that is able to increase saccharose content of glucoamylase capable of being produced an large amount by solid culture and enhance stability to heat and protease.
SOLUTION: This DNA has a base sequence that is shown by the formula and contains a promoter of glucoamylase genes isolated from Aspergilus orggae and/or a protein code domain. The transformant A (FERM P-15826) of this DNA is obtained by cloning cDNA that is considered to code glucoamylase expressed highly by solid culture and chromosome DNA fragments, coupling the former with an expression vector, transfecting Saccharomyces cerevisiae to obtain a transformant B (FERM P-15827), and transfacting the latter into an Aspergillus orggae host vector system.
