Abstract:

To obtain a new immortalized liver cell line, derived from human, which retains an ability of expressing genes coding for enzymes which are involved in the metabolism of xenobiotic substances in the liver, and can be used, for example, for the screening of compounds which inhibit or promote the activity of the enzymes.

This is a new immortalized liver cell line, from human, which retains an ability of expressing genes coding for enzymes, which are involved in the metabolism of xenobiotic substances in the liver, such as NADPH cytochrome P450 reductase, NADPH cytochrome P450, flavin monooxygenase, epoxyhydratase, glucuronyl transferase, sulfotransferase, and glutathione S- transferase, and can be used, for example, of the screening of compounds which inhibit or promote the activity of at least one of the enzymes or which inhibit or promote the expression of at least one of the genes coding for the enzymes. This cell line is obtained by introducing SV40T antigen gene into the liver parenchymal cell isolated from the liver from a dead human embryo.
