Abstract:
PURPOSE: To obtain a metal chelate resin suitable for purification of a protein, especially the purification of the protein containing adjacent histidine residues.
CONSTITUTION: This new nitrilotriacetic acid derivative is a compound of the formula H2N-(CH2)x-CH(COOH)-N(CH2COOH)2 [(x) is 2, 3 or 4] or a salt thereof. N-Terminus-protected compound of the formula R-HN-(CH2)x-CH(NH2)-COOH [R is an amino protective group; (x) is 2, 3 or 4] is reacted with two equivalents of bromoacetic acid and the protective group is eliminated from the product to provide the compound of the formula H2N-(CH2)x-CH(COOH)-N(CH2COOH)] in the method for preparing the nitrilotriacetic acid derivative.
