Abstract:
PURPOSE: To obtain the subject derivative for therapeutic agent, etc., for bone metabolic diseases such as osteoporosis, consisting of a steroid derivative containing a phosphoric acid group and capable of making a steroid compound to act on a bone structure more selectively compared with other organ.
CONSTITUTION: The objective new steroid derivative (salt) expressed by formula I [X-O-is steroid compound residue; A is formula II [(y) is 1-3; (p) is 0 or 1; (m) is 0-5; R1 is H, alkyl or aryl; Y is O or NH], etc.; R is H or alkyl] is obtained by reacting 17β-hydroxy-3-methoxymethyloxy-1,3,5-estratriene with N,N'-carbonylimidazole and reacting the reaction? product with a glycine ester and diazacycloundecene, further hydrolyzing the product and reacting the hydrolysis product with isobutyric acid chloride and successively, aminomethylenebisphosphosphonic acid tetraethyl ester, dissolving the reactio product in acetonitrile. adding water thereto while cooling and filtrating and drying the resultant precipitation.
