Abstract:

PURPOSE: To obtain a DNA useful for creating a plasminogen activating factor having potent activity to convert plasminogen into plasmin and useful as a thrombolytic agent.
CONSTITUTION: This DNA codes for an amino acid sequence of the formula (R is Ser, etc.; X is Lys, Ile, etc.; Y is Tyr-Ser-Gln-Pro-Gln-Phe-Arg-Ile, etc.; except that R is Ser, X is Lys, and Y is Tyr-Ser-Gin-Pro-Gin-Phe-Asp-Ile). The objective plasminogen activating factor is obtained by treating by conventional, means a complete DNA sequence or genome coding for natural or mutant t-PA transfected into a vector (e.g. pTPA21, pTPA25) obtained by partial or full DNA synthesis using a DNA synthesizer or from a transformant [e.g. E. coli LE392λ+ (pTPA21), E. coli JA221 (pTPA25) ATCC 39808]. The t-PA is active, well-reholdable, long in half-life and also excellent in thrombolytic activity.
