Abstract:
PROBLEM TO BE SOLVED: To obtain a new tricyclic compound having β3 operation activity, blood sugar lowering action and lipolytic activity, useful for treating and preventing diseases related to β3, such as diabetes, obesity and hyperlipemia.
SOLUTION: This compound is shown by formula I (R is H or methyl; R1 is H, a halogen, a hydroxyl group, benzyloxy, etc.; R2 is H, hydroxymethyl, nitro, etc.; R6 is H or a lower alkyl; X is a secondary nitrogen atom, an oxygen atom, a sulfur atom or methylene; when X is a secondary nitrogen atom, an oxygen atom or a sulfur atom, R9 is H, one of R7 and R8 is H and the other is H, amino, etc., and when X is methylene, R7 and R8 are H and R9 is H, amino, acetylamino or a hydroxyl group; *1 is an asymmetrical carbon atom; *2 is an asymmetric carbon atom when R6 is the lower alkyl) or its salt. The compound of formula I is obtained by reacting a compound of formula II (R1' is H, a halogen, etc.; R2' is H, nitro, etc.; A' is a hydroxyl-protecting group; B is Br or I) with a compound of formula III (Y is H; R7' to R9' are each are shown for R7 to R9) and deprotecting each protecting group.
