Abstract:

To provide a nonhuman animal having a higher-order brain functional disorder, deficient in a TAG-1 gene and suitably used for clarifying pathophysiological phenomena in the brain nervous system and diseases thereof, and to provide a method for producing the same.

This nonhuman animal having the higher-order brain functional disorder is deficient in a function of the TAG-1 gene on a chromosome. The nonhuman animal is produced as follows: the TAG-1 gene is isolated by screening genomic DNA libraries of the nonhuman animal; a transgene is built by using a homogeneous recombination vector the translation initiation region of which is substituted with a medicament selecting marker gene; the built transgene is introduced into an embryonic stem (ES) cell of the nonhuman animal by an electroporation technique; the ES cell in which the homogeneous recombination is caused by the medicament is selected by polymerase chain reaction and Southern blot techniques; a fertilized egg and the homogeneous recombination-caused ES cell are together mixed by an aggregation chimera technique so as to obtain a chimeric nonhuman animal; and such a reproductive chimeric nonhuman animal that the ES cell in a chimeric nonhuman animal individual is differentiated and formed into a reproductive cell is obtained.
