PURPOSE: To provide the novel enzyme useful for hydrolyzing cephalosporin C to produce aminocephalosporanic acid, and having a high enzymatic activity and a high processing effect.
CONSTITUTION: An enzyme of the formula [A1-268 is an amino acid sequence ranging from the Thr1 of natural cephalosporin C acylase (CCAase) to its Cly268; A272-304 is an amino acid sequence ranging from the Cln272 of CCAase to its Tyr; A306-773 is an amino acid sequence ranging from the Val306 of CCAase to its Ala773; X1-X3 are amino acids, but when X1 and X2 are Met and Ala, respectively, X3 is the groups excluding Cys]. The enzyme is obtained by a recombination DNA method comprising transforming a host cell with an expression vector recombined with a DNA coding the amino acid sequence, culturing the host cell, and subsequently separating the produced enzyme from the cultured product, or by a polypeptide synthetic method, etc.
