NEW MATERIAL: The (3S,4R)-isomer of a compound of formula I (the lactam and OH moieties are trans).
EXAMPLE: 3,4-Dihydro-2,2-dimethyl-6-ethenyl-4R-(2-oxo-piperidin-1-yl)-2H-1- benzopyran-3S-ol.
USE: The subject compound is useful for treatment of hypertension and/or respiratory tract disorders, because it exhibits hypotensive and respiratory tract dilating activities.
PROCESS: A compound of formula II (Ra is ethyl or a group or an atom convertible into ethyl) is reacted with a piperidinone anion of formula III or ammonia to give a trans-amino alcohol of formula IV. The product is acylated with a compound of the formula, L1(CH2)4COL2 (L1 and L2 are each a leaving group). When necessary, the product is cyclized and if Ra is other than ethyl, Ra in the product is converted into ethyl group to give the compound of formula I.
