PURPOSE: To produce a fatty acid acylated peptide having strong emulsifying activity, by preparing N-hydroxysuccinimide fatty acid acyl ester which is a novel substance, converting the compound to fatty acid acylated glycinine and decomposing the compound with a pronase.
CONSTITUTION: N-hydroxysuccinimide and palmitic acid are dissolved in tetrahydrofuran, dicyclohexylcarbodiimide is added to the solution to effect the reaction of the components and the residue is dissolved in ethyl acetate to obtain palmitic acid ester. The ester-containing tetrahydrofuran is added to a glycinine solution in a urea-containing buffer solution and the mixture is subjected to repeated centrifugal separation and dialysis to buffer solution to obtain palmitoylated glycinine. The glycinine is dissolved in a buffer solution, reacted in the presence of pronase E and extracted with a chloroform-methanol- butanol mixture to obtain the objective palmitoylated peptide as the decomposition product.
