NEW MATERIAL:A lymphokine (molecular weight, 15,000; chemically stable in acidic range; active to induce lymphotoxin-production of lymphocyte; devoid of interleukin-2 activity and interferon activity).
USE: Non-antibody substance of lymphotoxin-production inducing factor.
PREPARATION: A human T-cell hybridoma (e.g. KC8-1, KC8-1-9, KC8-1-10, etc.) prepared from a human T cell (human peripheral blood lymphocyte or lymphocyte separated from spleen or thymus gland) treated with the hemocyanin of chinese keyhole limpet (Macroschisma sinensis), is cultured, and the supernatant liquid of the culture product is concentrated by saturating with 90% ammonium sulfate. The objective refined lymphokine is obtained by passing the concentrate through a blue sephalose column, eluting with PBS, and collecting a specific fraction using the lymphotoxin production inducing activity as an index.
