Abstract:

PURPOSE: To obtain protoplast of bacterium, without pretreatment with an agent such as antibiotic, etc. during culturing, only by treating lysozyme-sensitive bacteria belonging to Corynebacterium genus or Brevibacterium genus with lysozyme.

CONSTITUTION: A lysozyme-insensitive strain belonging to Corynebacterium genus or Brevibacterium genus is subjected to the mutagenic treatment, and a strain which cannot be grown in a medium containing lysozyme at an extremely low concentration (<about 25μg/ml) to allow the complete growth of the parent strain and nevertheless can be grown in a lysozyme-free medium. The objective lysozyme-sensitive bacteria belonging to Corynebacterium genus or Breviacterium genus [e.g. Corynebacterium glutamicum, L-15 (FERM-P No.5946), Corynebacterium herculis L-103 (FERM-P No.5947), brevibacterium devaricatum L-204 (FERM-P No.5948), Brevibacterium lactofermentum L-312 (FERM-P No.5949), etc.] can be obtained by this process.
