Abstract:

PURPOSE: To obtain an antitumor polypeptide, consisting of a special amino acid sequence and having improved specific activity and stability, reaction spectrum extended into a wide region with reduced side effects.

CONSTITUTION: The aimed polypeptide expressed by the formula. This polypeptide can be produced by using a human TNF gene designed for using a human TNF precursor cDNA structural genetic part reported by Pennica et al. as a base, providing a cleavage part with a suitable restriction enzyme at suitable positions, respectively applying a translation starting codon ATG to the 5'-side and two translation end codons TGA and TAA to the 3'-side, providing a cleavage part with a restriction enzyme ClaI on the upstream side of the former, enabling linking of the SD sequence and the translation starting codon in a proper state to a promoter and providing a cleavage part with a restriction enzyme HindIII on the downstream side of the latter and capable of facilitating linking to a vector plasmid.
