PURPOSE: To improve the productivity of a physiologically active substance by culturing a specific microbial strain of genus Escherichia.
CONSTITUTION: An ATP is produced by linking a DNA containing a gene coding GSH-I, a DNA containing a gene coding GSH-II and a gene coding acetate kinase(ACK) to different vector DNAs. A DNA containing a gene coding an enzyme active to convert the above ATP into an ADP to synthesize a physiologically active substance is inserted into a bacteriophage vector to obtain a recombinant DNA. The DNA is enclosed with a coat protein of λ-bacteriophage using an in-vitro packaging process and the obtained bacteriophage is infected to a host cell. The infected cell strain is cultured on T-Y medium containing kanamycin to obtain a microorganism containing the above recombinant DNA and belonging to genus Escherichia capable of producing a physiologically active substance. The microorganism is inoculated on T-Y medium and cultured at 30-35°C for 2-24hr to obtain the objective physiologically active substance.
