Abstract:

To provide an oligonucleotide having an improved melting curve behavior, especially a FRET hybridization probe.

This method for synthesizing the oligonucleotide containing an axial molecular rod which has a rigid structure and can not bend by itself, comprises a process of providing a controlled hole glass particles and a process of synthesizing by a phosphoramidite chemical action. This oligonucleotide containing the axial molecular rod which has the rigid structure and can not bend by itself, in which the rod is covalently bonded at the 5' position of 5' terminal residue or at the 3' position of 3' terminal through a phosphate part, can be used as a primer for a primer elongation reaction or nucleic acid amplification reaction, and can also be used as a hybridization probe by which the temperature dependence of the hybridization can be monitored in real time.
