Abstract:
PURPOSE: To obtain an optically active trans-2-aryl-1-cyclohexanol derivative useful as an asymmetric source of asymmetric Diels-Alder reaction and a starting material of physiologically active substances.
CONSTITUTION: A compound of formula I (R1 is H, 1-15C alkyl, methoxymethyl, methoxyethyl, methylthiomethyl, tetrahydropyranyl, cyclopropylmethyl, allyl, cyclohexyl, benzyl, 9-anthorylmethyl or tert.-butyldimethylsilyl; R2 is H or 1-15C acyl; symbol * is asymmetric carbon atom), e.g. (1S, 2R)-trans-2-(2- hydroxyphenyl)-1-cyclohexanol. The compound is obtained by subjecting (±)- trans-2-aryl-1-cyclohexanol of formula II to transesterification with an aliphatic acid vinyl ester or triglyceride using an esterase (preferably derived from the genus Pseudomonas).
