Abstract:
PURPOSE: To provide new compounds showing an anti-inflammatory effect and useful as an anti-inflammatory agent, an antirheumatic agent, etc.
CONSTITUTION: Compounds of formula I (R1 is lower alkyl; R2 is H, lower alkyl or phenyl; R3 is H or lower alkyl; R4 and R5 are H, lower alkyl, lower alkylsulfonyl, formyl, etc.; A is heterocycle selected from isoxazolyl, oxazolyl and oxidiazolyl), e.g. 5-amino-3-[1-[4-(4-thiazolyl) phenyl]ethyl]isoxazole. The compound of formula I can be obtained by reacting a compound of formula II (R7 is lower alkyl) with a nitrile derivative of formula R8CH2CN (R8 is H or lower alkyl) and then reacting the resultant compound of formula III with a compound of formula NH2OH.HCL in the presence of an acid scavenger (e.g. pyridine).
