Abstract:

PURPOSE: To provide a novel compound useful as an antitumor agent, to be easily prepared as a pharmaceutical agent for having an improved water- solubility.

CONSTITUTION: A compound represented by formula I, (wherein T is a taxane portion having substituted 3-amino-2-hydroxypropanoyloxy on 13C carbon atom; m is 0 to 6; and n is 1 to 3), or a salt thereof, such as 7-O-(phosphonoxymethyl) paclitaxel is produced. The compound is produced by, for example, treating the formula: T'-(OH)n, (wherein T' is a taxane derivative with OH protected) of a paklitaxel represented by formula II, (wherein R is Ph; and R' is acetyl), or the like, with dimethyl sulfide and organic peroxide so as to obtain methylthioether represented by formula III, treating the compound with N- iodosuccinimide and phosphate represented by formula HOP(O)(ORy)2, (wherein Ry is a phosphono protecting group), and finally eliminating the phosphono protecting group and an optional OH protecting group from the obtained compound represented by formula IV.
