Abstract:
PURPOSE: To obtain a novel compound having a nucleotide transportation- inhibiting activity, and useful for protecting hearts from myocardiopathy caused by ischemia or anoxia.
CONSTITUTION: A compound of formula I (R1-R3 are H, lower alkyl, lower alkoxy, lower alkanoyl, lower alkoxycarbonyl, halogen, nitro, amino, etc.; R4 is H, halogen; X is H, carbamoyl, lower alkoxycarbonyl, etc.; (n) is 1-3; Ar is aryl. heterocyclic), e.g. N-(4-amino-2,6-dichlorophenyl)-2-carbamoyl-4-(6,7- dimethoxy-4-quinazolinyl)-1-piperazinyl-1-piperazine acetamide. The compound of formula I is obtained by reacting a compound of formula II (L is releasable group such as halogen, methanesulfonyloxy or toluenesul fonyloxy) with a compound of formula III in the presence of a base (e.g. triethylamine) in a solvent (e.g. methanol) at 0°C to the boiling point of the solvent for 5 minutes to 24 hours.
