Abstract:

PURPOSE: To provide a plasmid retainable stably in Bacillus brevis 47 capable of secreting proteins in a medium in an amount of as high as 12g/l under the optimum cultivation condition, and useful as a cloning vector of the strain.

CONSTITUTION: The objective plasmid has the restriction sites shown in the figure, and has Bacillus brevis as the host. It can impart the resistance to antibiotic substances or the productivity of protein to the whole plasmid or the region containing the replication initiation point, and to the host. The plasmid has been found in Bacillus brevis 481 (FERMP-7531) which is a relative strain of Bacillus brevis 47 (FERMP-7224), and was named as pWT481. Since the pWT481 is devoid of selective maker, it can be used for the introduction of erythromycin- resistant gene, e.g. by mixing with pHW1.
