Abstract:

PURPOSE: To make possible to carry out genetic engineering with one vector by constructing a plasmid vector containing drug resistance gene, promoter generated in Escherichia coli, repressor gene of promoter generated in Escherichia coli, etc.

CONSTITUTION: A plasmid vector containing drug resistance gene, promoter, generated in Escherichia coli, repressor gene of promoter generated in Escherichia coli, multicloning site, transprinting terminator, promoter of RNA polymerase and IG region of phage is synthesized as a mechanism of heredity. The concrete example is a plasmid vector PNUT-8 having specific restriction enzyme cleavage. Resultant vector is possible to carry out genetic engineering with one vector, without changing vector according to the object.
