PURPOSE: To provide an antibody the immunogen of which is a specified molecular species of rat liver cytochrome P-450. This antibody is useful, e.g. for estimation of the molecular specie of this enzyme exhibiting a catalytic effect on metabolism of a medicine by immunoblotting, quantitative analysis and study of distribution in the tissue according to the immune enzyme antibody method.
CONSTITUTION: When isolating and purifying a molecular species of rat liver cytochrome P-450, a methylcholanthrene-treated rat, a phenobarbital-treated rat, an ethanol- treated rat, a clofibrate-treated rat or a non-treated rat is used respectively in the case of CYP1A, in the case of CYP2B1, 2C6 or 3A2, in the case of CYP2E1, in the case of CYP4A1 or in the case of CYUP2C11 or 2C13. The liver is taken out from each rat and each molecular species of cytochrome P-450 is isolated and purified from the liver according to the conventional method. The resultant molecular species are subsequently each separately mixed with a adjuvant and intradermally injected to a goat, etc., to immunize the goat therewith and exsanguination is carried out when the antibody tier reaches a sufficient value. Serum separation is then carried out, thus affording the objective polyclonal antibody corresponding to each molecular species of rat liver cytochrome P-450.
